Overview

The T cell receptor (TCR) on HPV16-specific CD8+ T cells is a highly variable, heterodimeric membrane receptor (composed of α and β chains), specialized for recognizing short peptide fragments derived from the human papillomavirus type 16 (HPV16) presented on major histocompatibility complex class I (MHC-I) molecules. This receptor, in combination with the CD8 co-receptor, mediates antigen-specific activation of cytotoxic T lymphocytes, triggering targeted killing of HPV16-infected or malignant cells. Recognition depends on both the unique peptide specificity encoded in the TCR variable regions and the restriction by the presenting MHC allele. Such TCRs are critical for immune control of HPV16 infections and associated malignancies. In therapeutic contexts, genetically engineered TCRs targeting HPV16 antigens have been used in adoptive cell therapies for cancer treatment.
