Overview

T-cell receptors (TCRs) recognizing Blomia tropicalis allergen-derived epitopes are specialized heterodimeric surface proteins on T lymphocytes that play a pivotal role in the development of allergic sensitization to the tropical house dust mite. These receptors specifically bind to immunodominant epitopes from allergens such as Blo t 5, Blo t 7, and Blo t 21, which are presented by Major Histocompatibility Complex (MHC) class II molecules on antigen-presenting cells (APCs) [PMID: 25617545]. This recognition event is the primary trigger for the activation and differentiation of allergen-specific CD4+ T cells into a Th2 phenotype, leading to the production of IgE antibodies and the recruitment of inflammatory cells like eosinophils and mast cells [PMID: 30639434]. Consequently, these TCRs are central to the pathophysiology of allergic asthma and rhinitis in tropical and subtropical climates where Blomia tropicalis is prevalent. Therapeutic interventions, such as allergen-specific immunotherapy (AIT), target these TCR-mediated pathways to induce immune tolerance [PMID: 28213450]. By modulating TCR signaling through chronic exposure to low doses of allergen, AIT promotes the expansion of regulatory T cells (Tregs) and the production of IL-10, effectively dampening the allergic cascade.
