The T cell receptor (TCR) recognizing CLB-505-derived HBsAg peptides presented on MHC is a specialized immune receptor that mediates the cellular response against the Hepatitis B virus (HBV). CLB-505 is a modified Hepatitis B surface antigen (HBsAg) developed by ClearB Therapeutics, engineered to display specific epitopes associated with the functional cure of chronic hepatitis B (CHB) (ClearB Therapeutics, 2023). These epitopes, when processed and presented on Major Histocompatibility Complex (MHC) molecules, are recognized by specific TCRs on the surface of T cells, triggering activation, proliferation, and effector functions such as the secretion of antiviral cytokines like IFN-gamma and the direct lysis of infected hepatocytes (EASL Congress, 2022). In the context of therapeutic vaccines like CLB-3000 and CLB-4000, the goal is to induce or expand the population of T cells bearing these specific TCRs to achieve sustained viral control and HBsAg loss (BioWorld, 2023). This TCR-peptide-MHC interaction is a critical component of immunotherapies aiming for a functional cure of chronic HBV infection and the prevention of HBV-related hepatocellular carcinoma (AASLD, 2024).
The TCR binds to specific HBsAg epitopes derived from the CLB-505 antigen when presented on MHC molecules, triggering T cell activation and the subsequent elimination of HBV-infected hepatocytes.
