The T-cell receptor (TCR) recognizing Guanylate-binding protein 5 (GBP5)-derived peptides is an engineered or naturally occurring receptor designed to target cancer cells overexpressing the GBP5 protein. GBP5 is an interferon-inducible GTPase that plays a role in innate immunity and inflammasome assembly, but it is frequently upregulated in various malignancies, including glioblastoma, melanoma, and triple-negative breast cancer. When GBP5-derived peptides are processed and presented on the cell surface by Major Histocompatibility Complex (MHC) molecules, specifically HLA-A*02:01, they serve as tumor-associated antigens. TCR-engineered T-cell therapies (TCR-T) utilize these specific receptors to redirect the patient's immune system to recognize and eliminate GBP5-positive tumor cells. This approach leverages the high specificity of TCR-MHC interactions to provide a targeted immunotherapy option for cancers where GBP5 acts as a driver or marker of progression.
