Overview

The T cell receptor (TCR) recognizing IMA950 peptide–MHC complexes is a specialized immune receptor that mediates the recognition of tumor-associated antigens (TAAs) in glioblastoma. IMA950 is a multi-peptide vaccine developed by Immatics, consisting of 11 tumor-associated peptides (TUMAPs) identified through mass spectrometry of HLA-presented peptides on primary glioblastoma tissue, including antigens such as PTPRZ1, BCAN, and MET. These peptides are presented by HLA-A*02 (9 peptides) or HLA-DR (2 peptides) molecules on the surface of tumor cells and antigen-presenting cells. The specific TCRs that recognize these peptide-MHC (pMHC) complexes are the primary targets for activation by the IMA950 vaccine. Upon binding to their cognate pMHC ligands, these TCRs trigger T cell activation, proliferation, and cytotoxic activity against the tumor. This interaction is essential for the vaccine's ability to induce a broad, multi-clonal anti-tumor immune response involving both CD8+ and CD4+ T cells. Therapeutic strategies involving this target include the use of IMA950 in combination with adjuvants like poly-ICLC and GM-CSF, as well as immune checkpoint inhibitors like pembrolizumab. Safety concerns associated with targeting these receptors in the brain include the risk of cerebral edema due to localized inflammation.
