The T cell receptor (TCR) recognizing H1-derived peptides in the context of MHC class II is a critical component of the adaptive immune response against Influenza A virus, specifically the H1 subtype. These TCRs are primarily expressed on CD4+ T helper cells and recognize peptide fragments derived from the viral hemagglutinin (HA) protein presented by Major Histocompatibility Complex (MHC) class II molecules, such as HLA-DR or HLA-DQ. This recognition event is essential for 'linked recognition,' a process where H1-specific T cells provide the necessary signals, including CD40 ligand and cytokines, to B cells to promote high-affinity antibody production and the establishment of long-term humoral immunity. In the context of drug development, these TCRs are primary targets for universal influenza vaccines designed to elicit broad, cross-reactive T cell help across different viral strains. They are also being explored as therapeutic agents in TCR-engineered T cell (TCR-T) therapies for severe viral infections. However, these TCRs are associated with significant safety considerations, most notably molecular mimicry between H1 peptides and self-antigens like hypocretin, which has been implicated in the development of narcolepsy following certain H1N1 vaccinations.
