Overview

T-cell receptors (TCRs) recognizing IPX peptide–HLA complexes are specialized immune receptors engineered or selected to target specific intracellular antigens presented on the cell surface. The IPX peptide is a tumor-associated or pathogen-derived antigen that, when presented by Human Leukocyte Antigen (HLA) molecules (most commonly HLA-A*02:01), serves as a unique signature for diseased cells. These TCRs are the core component of TCR-T cell therapies, where a patient's T-cells are modified to express the specific receptor, enabling them to identify and eliminate cells expressing the IPX antigen. This approach is particularly valuable for targeting intracellular proteins that are not accessible to traditional antibody-based therapies. In clinical development, these receptors are being evaluated for their efficacy in treating various solid tumors and infectious diseases, with a focus on achieving high specificity to minimize off-target effects.

Mechanism of action

Engineered T-cells expressing these receptors bind specifically to the IPX peptide presented by HLA molecules on the surface of target cells, leading to T-cell activation and targeted cell lysis.
