Overview

This target is a T-cell receptor (TCR) engineered or naturally occurring to specifically recognize a peptide derived from tyrosinase, a melanocyte differentiation antigen, when presented by the HLA-A2 major histocompatibility complex (MHC) class I molecule. Targeting the tyrosinase-HLA-A2 complex allows for the immune recognition and destruction of melanoma cells expressing both these antigens. Some engineered TCRs, such as TIL1383I, have been structurally characterized and demonstrate atypical geometry and CD8-independence, which may affect their clinical utility and safety. TCR-based therapies targeting this epitope are in clinical development for adoptive cell transfer approaches in cancer immunotherapy.
