The T-cell receptor specific for brachyury peptide-major histocompatibility complex is a membrane-bound immune receptor found on the surface of T lymphocytes, engineered or naturally occurring, which binds specifically to a peptide derived from the transcription factor brachyury presented by a specific MHC/HLA molecule (commonly HLA-A2) on target cells. This interaction is essential for targeted immunotherapies in cancer, enabling selective recognition and killing of tumor cells expressing the brachyury antigen. The TCR-peptide-MHC interaction is highly specific, allowing the immune system (or engineered T cells) to discriminate between healthy and tumor cells based on their antigen presentation profile. Such approaches form the basis for adoptive T cell therapies and peptide vaccine strategies targeting cancers with elevated brachyury expression.
Recognition of brachyury-derived peptide presented by MHC (HLA-A2) on tumor cells by the T-cell receptor leads to activation of T cells, release of cytotoxic granules, and target cell death. MHC restriction ensures killing only of tumor cells presenting the specific peptide-MHC complex.
