T-cell-specific surface glycoprotein CD3 is a multi-subunit protein complex expressed on the surface of all mature T cells and serves as both a co-receptor and signaling apparatus for the T-cell receptor (TCR). Comprised of CD3γ, CD3δ, two CD3ε, and CD3ζ chains, the complex is essential for T-cell activation: upon antigen recognition by the TCR, CD3 transduces intracellular signals via immunoreceptor tyrosine-based activation motifs (ITAMs) in its cytoplasmic tails. The CD3 complex is the main therapeutic target for monoclonal antibodies modulating T-cell activity (e.g., muromonab-CD3) and is pivotal not only in immune defense against pathogens, but also in pathological processes including cancer, autoimmunity, and transplantation. Its structural integrity, mechanism of signal transduction, and role in orchestrating immune responses have made it a major focus in immunology and drug development.
