Overview

CD3 is a multimeric protein complex associated with the T cell receptor (TCR) on T lymphocytes. It is essential for TCR assembly, surface expression, and signal transduction following antigen recognition. The complex consists of CD3εγ, CD3εδ, and CD3ζζ dimers. Upon antigen binding to the TCR, ITAM motifs within the CD3 cytoplasmic tails are phosphorylated, initiating downstream signaling cascades that lead to T cell activation. CD3 is expressed on all mature T cells and is used as a marker for identifying and quantifying T cells. Anti-CD3 antibodies are used therapeutically to modulate immune responses, such as in transplant rejection.
