The CD3 complex is a multi-subunit protein complex on T lymphocytes, crucial for T cell development, antigen recognition, and signal transduction. It consists of CD3E, CD3G, CD3D, and CD3Z subunits forming heterodimers (CD3δε, CD3γε) and homodimer (CD3ζζ), associated with the TCR αβ or γδ heterodimers. ITAMs in the cytoplasmic domains initiate signaling upon antigen recognition. Defects can cause severe combined immunodeficiency and autoimmunity.
Antibodies targeting CD3 can deplete T cells or modulate T cell activity. Small molecules can inhibit signaling pathways downstream of CD3 activation.
