The T cell surface glycoprotein CD3 complex chain delta (CD3δ) is one of four invariant chains (γ, δ, ε, ζ) of the CD3 complex, which is essential for surface expression and signal transduction of the T cell receptor (TCR) complex. The CD3 complex stabilizes the TCR on the surface of all mature T cells and transduces signals following antigen recognition. CD3δ forms a dimer with CD3ε as part of the functional CD3 structure, enabling the transmission of activation signals via immunoreceptor tyrosine-based activation motifs (ITAMs) in their cytoplasmic domains. CD3δ is required for normal T cell development, activation, and effector function; mutations or loss of CD3δ expression results in immunodeficiency. The CD3 complex, including the delta chain, is a major target for monoclonal antibody therapies in transplantation, autoimmune diseases, and cancer immunotherapy.
Monoclonal antibodies against CD3 trigger T cell depletion, activation, or anergy by binding to the CD3 complex, leading to signaling dysregulation, cell death, or immune modulation. Agonism or antagonism of the TCR/CD3 complex for immunosuppression or immunostimulation.
