Overview

T cell surface glycoprotein CD3 epsilon is a transmembrane protein that is an essential component of the CD3 complex, which, together with the T cell receptor (TCR) α/β or γ/δ heterodimers, forms the TCR–CD3 complex critical for T cell development, activation, and immune signaling[3][4][7]. CD3ε is found in two distinct heterodimers (CD3γε and CD3δε) that noncovalently assemble with TCR on the T cell surface[4][7]. Its cytoplasmic tail contains immunoreceptor tyrosine-based activation motifs (ITAMs), which are phosphorylated upon antigen engagement, initiating downstream signaling needed for T cell immune functions[1][3]. CD3ε can be targeted therapeutically by specific monoclonal antibodies, relevant in autoimmune diseases, transplant rejection, and T cell malignancies[2][3]. It also serves as a robust biomarker for the identification of T cells in clinical and research settings.
