CD5 is a 67 kDa type I transmembrane glycoprotein and a member of the scavenger receptor cysteine-rich (SRCR) superfamily. It is constitutively expressed on all mature T cells and thymocyte precursors and also on the B-1a subset in mice and some regulatory/anergic human/mouse B-cells. CD5 acts primarily as a negative regulator of antigen receptor signaling, inhibiting T-cell receptor (TCR) and B cell receptor (BCR) signaling and promoting T cell nonresponsiveness and survival. Modulation of CD5 within the tumor microenvironment corresponds to a strategy adopted by tumor-specific cytotoxic T lymphocytes to optimize their cytotoxic and cytokine secretion functions.
