Overview

T cell surface markers used for lipid nanoparticle-mediated uptake refer to a collection of proteins expressed on the plasma membrane of T lymphocytes that are targeted to facilitate the entry of lipid nanoparticles (LNPs). These markers, including CD3, CD4, CD8, and CD5, are utilized by decorating LNPs with specific ligands like monoclonal antibodies or single-chain variable fragments (scFvs) that bind the receptor and trigger receptor-mediated endocytosis (Rurik et al., Science, 2022). This approach is central to the development of in vivo CAR-T cell therapies, where mRNA-carrying LNPs are used to transiently or permanently reprogram T cells within the circulatory system to target specific diseases (Tombácz et al., Molecular Therapy, 2021). Beyond oncology, these markers are being investigated for treating cardiac fibrosis and autoimmune disorders by delivering modulatory genetic material (Capstan Therapeutics, 2023). While promising, the use of these markers requires high specificity to avoid off-target delivery to the liver or other immune cell subsets, and to prevent premature T-cell activation or exhaustion (Sago et al., PNAS, 2018).
