Overview

T-complex protein 11 homolog (TCP11) is a testis-specific protein crucial for male fertility, particularly for the processes of spermatogenesis, sperm maturation, and motility[1][2][3]. It functions as the putative receptor for fertilization-promoting peptide (FPP)[1][3]. In both mouse and human, TCP11 is highly expressed in the testis—with variant expression in tissues such as brain and epididymis in humans—and localizes to structures such as the acrosomal vesicle and sperm flagellum[1][2][3]. TCP11 is required for sperm tail structural integrity and motility, and its deficiency is associated with morphological abnormalities and impaired sperm function, leading to male infertility[1][2][3]. TCP11 appears to regulate sperm capacitation, potentially via the cAMP/PKA signal transduction pathway[2][3]. Recent evidence suggests TCP11 may also play a role in cancers such as cervical cancer, where altered expression affects cellular proliferation and apoptosis[1]. However, its major characterized function is in germ cell development and male fertility. TCP11 is not established as a therapeutic target, biomarker, or druggable protein in current clinical use[1][2][3].
