The T helper 1 (Th1) cell response is a fundamental component of the adaptive immune system, characterized by the differentiation of CD4+ T cells into Th1 effectors primarily under the influence of interleukin-12 (IL-12) and the master transcription factor T-bet [StatPearls, 2023]. This response is essential for cell-mediated immunity, orchestrating the defense against intracellular pathogens such as viruses and mycobacteria by activating macrophages and enhancing the activity of CD8+ cytotoxic T cells [NCBI, 2022]. Consequently, the Th1 pathway is a major focus of pharmacological intervention, with therapies designed to either suppress excessive Th1 activity in autoimmune diseases using TNF inhibitors and IL-12 blockers or to stimulate the response in the context of cancer immunotherapy and chronic infections [Frontiers in Immunology, 2020].
Modulation of the Th1 response typically involves the neutralization of effector cytokines (e.g., TNF-alpha, IFN-gamma) or the inhibition of upstream signaling molecules like IL-12 and JAK/STAT proteins that drive Th1 differentiation and maintenance [Frontiers in Immunology, 2020].
