T helper 1 (Th1) cell response induction refers to the process by which naïve CD4+ T cells differentiate into Th1 effector cells. This differentiation is a critical component of the adaptive immune system, primarily responsible for orchestrating cell-mediated immunity against intracellular pathogens such as viruses and certain bacteria. The induction of a Th1 response involves specific cytokines, transcription factors, co-stimulatory molecules, and chemokine receptors that collectively guide the fate and function of these immune cells. Key cytokines include IL-12 and IFN-γ. Key transcription factors include STAT4, STAT1 and T-bet. Co-stimulatory molecules like CD28, ICOS, CD226 and Tim3 play modulatory roles. Chemokine receptor CXCR3 guides T cell movement during early stages of Th1 development. Once differentiated, Th1 cells activate macrophages, promote cytotoxic responses, and support IgG class switching.
