Overview

CD4-positive T cell differentiation is the biological process by which naïve CD4+ T lymphocytes specialize into various effector and regulatory lineages upon antigen stimulation and exposure to a complex cytokine environment. Major subtypes include Th1, Th2, Th17, regulatory T cell (Treg), follicular helper T cell (Tfh), Th9, and Th22 cells, each defined by unique transcription factors and cytokine production profiles. These differentiated subsets orchestrate distinct immune responses, play pivotal roles in infection control, malignancy, autoimmunity, and inflammation, and are regulated through integrated signals from antigen-presenting cells, cytokines, and cell surface receptors. Targeting elements of this process (e.g., cytokines, transcription factors) is a therapeutic strategy in diverse diseases, but modulation of CD4+ T cell differentiation as a whole is not itself directly druggable[1][2][3][5].
