T helper (Th) cell differentiation via cytokine induction is the process by which naïve CD4+ T cells develop into distinct functional subsets in response to specific cytokines present in their environment. This differentiation is crucial for orchestrating adaptive immune responses, as each Th subset specializes in different aspects of immunity and inflammation. Key subsets include Th1, Th2, Th17, Tfh, and iTreg, each characterized by unique inducing cytokines, transcription factors, and effector functions.
Cytokines bind to their receptors on naïve CD4+ T cells, activating downstream signaling pathways (primarily JAK/STAT) that induce the expression of lineage-specific transcription factors (e.g., T-bet, GATA3, RORγt, FoxP3, Bcl6), leading to differentiation into distinct Th subsets.
