Overview

T helper type 2 (Th2) cells are a subset of CD4+ T lymphocytes characterized by their ability to secrete signature “type 2” effector cytokines including interleukin‑4, interleukin‑5, and interleukin‑13. These cells play critical roles in orchestrating humoral immunity against extracellular parasites like helminths and contribute centrally to allergic inflammatory diseases such as asthma and atopic dermatitis. The differentiation into Th2 cells is driven primarily by exposure to IL–4 via STAT6 activation and upregulation of GATA3 transcription factor. Upon antigen recognition in peripheral tissues, differentiated effector Th2 cells secrete their characteristic set of pro-inflammatory mediators which promote B-cell class switching to IgE production, recruit eosinophils/basophils/mast cells, and drive tissue responses associated with allergy[1][3][4].
