T lymphocyte activation via ADCC enhancement

01

Overview

Activation of T lymphocytes, particularly CD4+ T cells, enhances antibody-dependent cellular cytotoxicity (ADCC) by promoting NK cell activation and function via cytokine (e.g., IL-2) secretion[6][7]. This approach can overcome therapeutic resistance in cancer by engaging T cells to improve the efficacy of monoclonal antibodies, such as anti-HER2 and anti-CD20 agents. Mechanistically, T cell help augments the cytotoxic activity of NK cells and other ADCC effectors, supporting tumor regression and improved clinical outcomes in cancer patients[6][7]. Drug strategies include bispecific antibodies that target tumor antigens and activate T cells simultaneously[6].

Mechanism of action

T cell activation (especially CD4+ T cells); Production of interleukin-2 (IL-2) by T cells, promotes NK cell functionality and ADCC capacity; Bispecific antibodies (e.g., HER2 × CD3) co-engage tumor antigen and T cells, amplifying ADCC.
