T lymphocyte activation via costimulatory molecule signaling is the process by which T cells require a secondary, antigen-independent signal, in addition to T cell receptor (TCR) engagement, to enable full activation, proliferation, and effector function[3][5]. These costimulatory signals are mediated by various receptor-ligand pairs, notably CD28 binding to B7 molecules (CD80/CD86) on antigen-presenting cells, among many other pairs such as ICOS/ICOS-L, 4-1BB/4-1BBL, and OX40/OX40L[1][3][5]. This process is critical for proper immune responses, prevention of autoimmunity, and is a central focus in immunotherapy for cancer, autoimmunity, and transplant rejection[1][4][5].
