Overview

Tachykinin precursor 3 (TAC3) encodes a secreted neuropeptide precursor, which is processed to generate Neurokinin B, a key member of the tachykinin peptide family. Neurokinin B is highly expressed in the central nervous system and acts primarily as a neurotransmitter and regulator of smooth muscle physiology. It binds the neurokinin-3 receptor, a G protein-coupled receptor, initiating diverse signaling pathways that control neurological, endocrine, immune, and vascular functions. TAC3 has important roles in reproductive endocrinology, particularly in the regulation of gonadotropin release, and is associated with diseases such as hypogonadotropic hypogonadism and infertility. Dysregulation or mutation of TAC3 can impact neurological, cardiovascular, and reproductive physiology, and it is considered an investigational target for multiple therapeutic applications, including reproductive medicine and neuropsychiatric disorders[1][2][5].

Mechanism of action

Antagonists inhibit NK3 receptor activation, modulating central and peripheral effects of Neurokinin B, which include reduction of gonadotropin secretion, modulation of vasomotor symptoms (hot flashes) in menopause, and other cardiovascular/neurophysiological effects. Inhibition of GPCR signaling via NK3 receptor reduces downstream effects (Ca²⁺ mobilization, PKC, cAMP, inflammatory lipid mediator synthesis).
