Overview

Talin rod domain-containing protein 1 (TLNRD1) is an α-helical actin-binding protein structurally homologous to the talin R7R8 rod domain, capable of binding and bundling F-actin through dimerization[1][5]. It participates in the promotion of filopodia formation and regulates cell proliferation, migration, and invasion, especially in cancer cells[1][2][5]. TLNRD1 is thought to possess potential oncogenic functions and its expression is regulated by microRNA (such as miR-574-3)[2][3][4]. It also interacts with other multidomain proteins containing LD motifs, such as RIAM and KANK, indicating a role in actin cytoskeleton regulation and cell motility[1]. While associated with cancer and cellular migration processes, there are no established drugs directly targeting TLNRD1 nor is it an established therapeutic target in current practice[4][5].
