Target intelligence / Profile preview

TAM family receptors (TAM)

Target
TAM
Molecular classification
Receptor, Receptor tyrosine kinases (RTKs)
01

Overview

The TAM family receptors (Tyro3, Axl, Mer) are receptor tyrosine kinases that play critical roles in immune homeostasis, efferocytosis, hemostasis, and cell survival/proliferation. They are activated by ligands Gas6 and Protein S, which bind to their extracellular Ig-like domains, leading to receptor dimerization and activation of intracellular signaling pathways. Dysregulation of TAM receptors is implicated in various diseases, including autoimmune disorders, inflammation, thrombosis, and cancer.

02

Mechanism of action

Ligand binding (Gas6, Protein S) leading to receptor dimerization, autophosphorylation, and downstream signaling cascades.

Safety considerations

Dual role in cancer (tumor growth regulation and anti-tumor immunity modulation) requires careful consideration in therapeutic targeting.
