Overview

Tandem C2 domains nuclear protein (TC2N) is a protein characterized by having two C-terminal C2 domains, which are involved in calcium-dependent phospholipid binding, clathrin binding, and protein–protein interactions[1][4][5]. TC2N is localized to the nucleus and has been implicated in mediating various cellular processes, including signal transduction, regulation of cell proliferation, and apoptosis. It is recognized as a context-dependent regulator with oncogenic properties in lung and gastric cancers, where it promotes cell growth, migration, and survival, in part by inhibiting the p53 tumor suppressor pathway through enhancement of CDK5 degradation and disruption of CDK5–p53 interactions[1][2]. In breast cancer, TC2N may act as a tumor suppressor via inhibition of the PI3K/AKT signaling pathway[1]. The gene is found on human chromosome 14q32. TC2N is being actively studied as a biomarker and potential therapeutic target in oncology, although there are currently no specific drugs directly targeting it[1][2][5].
