Serine/threonine-protein kinase TBK1 (Tank-binding kinase 1, TBK1) is a critical enzyme involved in the regulation of innate immunity, inflammation, and several other cellular processes. It mediates signaling downstream of pattern recognition receptors such as Toll-like receptors (TLRs), RIG-I-like receptors (RLRs), and STING-mediated cytosolic DNA sensors. TBK1 phosphorylates interferon regulatory factors IRF3 and IRF7, initiating transcription of type I interferon genes and other pro-inflammatory cytokines. Beyond antiviral defense, TBK1 regulates cell proliferation, apoptosis, autophagy, energy homeostasis, and anti-tumor immunity. Dysregulation can contribute to autoimmune diseases, neurodegeneration, or cancer development.
