Tankyrase refers to a family of multifunctional enzymes, specifically poly(ADP-ribose) polymerases (PARPs), that play critical roles in several cellular processes. In humans, the two main tankyrases are Tankyrase 1 (TNKS1) and Tankyrase 2 (TNKS2). These enzymes catalyze poly(ADP-ribosyl)ation (PARylation), regulating telomere homeostasis, Wnt/β-catenin signaling, cell cycle progression, and other cellular functions. Tankyrases have been implicated in cancer and other diseases and are a target for small molecule inhibitors primarily developed for cancer therapy.
Tankyrase inhibitors suppress Wnt/β-catenin signaling by stabilizing AXIN proteins, leading to reduced cell proliferation and differentiation.
