Overview

TAP binding protein-like (TAPBPL) is a cell-surface glycoprotein and member of the immunoglobulin superfamily that is structurally related to the B7 family. Expressed on T cells, antigen-presenting cells (B cells, monocytes, macrophages, dendritic cells), and some tumor tissues, TAPBPL functions as a negative regulator of T cell responses. It binds to MHC class I complexes and modulates antigen processing and presentation. TAPBPL suppresses T cell activation and proliferation; a soluble TAPBPL-Fc fusion protein has been shown to reduce autoimmune disease severity in mice, while blocking its activity with antibodies enhances antitumor immune responses. Thus, TAPBPL represents a novel immune checkpoint with potential implications in cancer immunotherapy, autoimmune disease, and infection
