Target of Myb1 membrane trafficking protein (TOM1) is a cytosolic adapter protein involved in intracellular membrane trafficking of ubiquitinated proteins, serving critical roles in autophagy, endosomal maturation, receptor recycling, and ubiquitin-dependent signaling. TOM1 contains a VHS domain and a GAT domain, supporting its ability to bind ubiquitin and clathrin, and facilitates the recruitment of ubiquitinated cargo onto early endosomes via interactions with proteins such as TOLLIP, MYO6, and ZFYVE16. In humans, TOM1 is associated with immune system function, including the inhibition of Toll-like receptor signaling and immune receptor recycling. While disruption of TOM1 is linked to primary immunodeficiency syndromes, there are no approved drugs or well-established biomarkers involving TOM1 in clinical practice. TOM1 is also an essential factor for the multiplication of certain plant viruses but in mammals is not a direct drug target or major receptor/enzyme family member[1][3][4][7].
