TATA-box binding protein associated factor, RNA polymerase I subunit D (TAF1D), is a component of the SL1/TIF-IB complex required for RNA polymerase I transcription initiation and ribosomal RNA synthesis. TAF1D localizes to the nucleolus and is essential for the assembly of the preinitiation complex at rDNA promoters. Loss or downregulation of TAF1D impairs SL1 complex recruitment, reduces Pol I transcription, inhibits cell proliferation, and induces cell cycle arrest. TAF1D has emerged as a biomarker of poor prognosis in several cancers, particularly osteosarcoma and MYCN-amplified neuroblastoma, where it regulates G2/M phase-related genes. As such, TAF1D is considered a promising therapeutic target in malignancies associated with altered ribosome biogenesis or cell cycle control
Inhibition of TAF1D reduces RNA polymerase I recruitment and rRNA synthesis, leading to cell cycle arrest (notably at G2/M) and reduced cell proliferation
