TATA-box binding protein associated factor 11 (TAF11) is a small, evolutionarily conserved subunit of the general transcription factor IID (TFIID) complex, which is essential for transcription initiation by RNA polymerase II[1][2][3]. TAF11 interacts directly with the TATA-box binding protein (TBP) and with TAF13, forming a histone-fold heterodimer that binds the concave DNA-binding surface of TBP and regulates TBP’s access to core promoter DNA[1][2]. This complex formation is mutually exclusive with TATA-box DNA binding, serving as a regulatory mechanism for TFIID assembly and promoter selection[2]. TAF11 plays critical roles in assembly of the preinitiation complex, promoter recognition, and the regulation of eukaryotic gene expression, and is essential for normal cell growth and viability[1][2][3]. There are no known approved drugs targeting TAF11 and it is not classified as a druggable therapeutic target.
