Overview

TATA-box binding protein associated factor 12 (TAF12) is a highly conserved subunit of the transcription factor IID (TFIID) complex, which is critical for the initiation of transcription by RNA polymerase II[2][3]. TAF12 interacts with the TATA-binding protein (TBP) and other TAFs, notably forming a histone-like heterodimer with TAF4[1][3]. Through these interactions, TAF12 facilitates assembly of the transcription pre-initiation complex and contributes to regulation of gene expression across eukaryotes[2][3]. TAF12 may also participate in additional regulatory complexes such as SAGA, and its precise requirement and function can vary in different cell types and organisms[1]. Disruption of TAF12 function can impair cell growth and viability, reflecting its central role in gene regulation. There are reported associations with certain cancers, but TAF12 is not currently a direct drug target and there are no approved therapeutic strategies targeting this factor[2].
