Overview

TATA-box binding protein associated factor 5 like (TAF5L) is a WD-repeat protein that forms a core component of multiprotein complexes involved in transcriptional regulation, including the human PCAF histone acetylase complex, which is analogous to the yeast SAGA complex[1][2][3][10]. As part of these complexes, TAF5L is essential for efficient histone acetylation, promoter recognition, transcription initiation by RNA polymerase II, and epigenetic control of gene expression[3][10]. TAF5L is required for myogenic transcription and cellular differentiation, acting as a coactivator and interacting with transcription factors such as TAF9 and SPT3 homolog[1][3]. The protein, encoded by the TAF5L gene on human chromosome 1, is structurally similar to other histone-like TAF proteins and exists in several alternatively spliced isoforms[1][3]. TAF5L is not considered a direct therapeutic target such as a receptor or enzyme, has no known approved drugs or established clinical biomarkers, and carries no specific safety concerns reported in the current literature[3][10].
