Overview

**TATA box-binding protein-like 1 (TBPL1)** is a member of the TATA box-binding protein family and is encoded by the TBPL1 gene in humans. Unlike the canonical TATA-binding protein (TBP), TBPL1 does not bind the TATA box sequence but can initiate transcription from TATA-less promoters[2]. It plays a key role in transcriptional regulation, especially for genes lacking a canonical TATA box. TBPL1 is essential in spermatogenesis and may be important for the expression of certain developmentally regulated genes[1][2]. While it shares homology with TBP, TBPL1 serves specialized functions and does not bind to the same promoter elements as TBP or associate with the full TFIID complex. Currently, TBPL1 is not considered a therapeutic target, nor are there known drugs or established biomarkers directly linked to its function or modulation[2][1]. **Note:** - TBPL1 is distinct from TATA-box binding protein (TBP) and other TBP-like factors such as TRF3, TRF1, and TRF2[4]. - Interactions include binding to other general transcription factors, such as GTF2A1[1]. - The gene has alternate transcripts and a pseudogene on chromosome 3[2].
