TATA-box binding protein-like 2 (TBPL2, also known as TBP-related factor 3/TRF3) is a vertebrate-specific member of the TATA-binding protein family that participates in transcription initiation by binding to the TATA-box motif in gene promoters and interacting with core transcription machinery such as TFIIA and TFIIB[1][2][4]. TBPL2 is most highly expressed in oocytes, where it replaces conventional TBP to support oocyte-specific transcription programs necessary for folliculogenesis and fertility. The protein is highly homologous with TBP in its C-terminal DNA-binding domain (95% identity), but its N-terminal domain is divergent, allowing specialization for oocyte function[1][2][4]. Loss of TBPL2 does not cause gross developmental abnormalities but leads to infertility in female mice due to defective folliculogenesis[1]. TBPL2 is not known to be a direct drug target in current therapeutic contexts.
