Overview

Tau protein acetylated at lysine 280 (K280) is a post-translationally modified form of the microtubule-associated protein tau. Acetylation at K280 disrupts normal tau function by reducing microtubule binding and increasing aggregation, promoting toxic gains-of-function and prion-like propagation. It is specifically detected in neuronal and glial inclusions characteristic of several "4R-tau" tauopathies, including Alzheimer’s disease. Monoclonal antibodies like Y01 and ADEL-Y01 are under development to target this epitope.

Mechanism of action

Monoclonal antibodies selectively bind to acK280-modified tau, blocking its propagation, aggregation, and secretion; promoting phagocytosis by microglia and improving neuronal viability.
