TBC1 domain family member 10A (TBC1D10A) is a cytoplasmic GTPase-activating protein that specifically inactivates RAB27A and RAB35 by accelerating the hydrolysis of their bound GTP. It is implicated in regulating intracellular vesicle trafficking, endosomal recycling, and proteolytic pathways. TBC1D10A contains a TBC (Tre-2/Bub2/Cdc16) domain, which confers its GAP activity. It also can bind PDZ domains and interact with cytoskeletal elements, but is not a receptor, transporter, ion channel, or transcription factor. Genetic variants in TBC1D10A have been associated with rare syndromes (e.g., Warburg Micro Syndrome 4), but there is currently no evidence that it is a direct therapeutic target or clinical biomarker
