Overview

TBC1 domain family member 15 (TBC1D15) is a cytosolic protein containing a conserved TBC (Tre-2/Bub2/Cdc16) domain and functions as a GTPase-activating protein (GAP) for Rab GTPases, primarily Rab7, with weaker activity on Rab11[1][3][4]. Its major role is to promote the conversion of active Rab7-GTP to inactive Rab7-GDP, thereby regulating late endosomal trafficking and vesicle transport[3][4]. TBC1D15 is ubiquitously expressed and evolutionarily conserved, with structural similarity to yeast Ypt/Rab-GAPs such as Gyp7p[1][3]. Mutations or altered expression of TBC1D15 have been associated with neurodegenerative and developmental diseases, specifically Charcot-Marie-Tooth disease type 2B and Warburg Micro syndrome 4[4]. There is also growing evidence that members of the TBC1 domain family play roles in cancer metabolism and prognosis, although direct mechanisms and therapeutic implications for TBC1D15 remain under investigation[5].
