TBC1 domain family member 16 (TBC1D16) is a cytosolic enzyme classified as a Rab GTPase-activating protein, characterized by a conserved TBC (Tre2/Bub2/Cdc16) domain and serine-rich C-terminal region. TBC1D16 regulates intracellular trafficking by acting as a negative regulator of specific Rab GTPases (notably Rab4A and Rab5C), modulating receptor recycling and degradation for proteins such as EGFR and the transferrin receptor. TBC1D16 functions as a host restriction factor against certain viruses (e.g., prototype foamy virus), inhibiting viral replication, and promoting type I interferon responses. In cancer, particularly melanoma, aberrant activation of TBC1D16 (e.g., the 47 kDa isoform) contributes to tumor growth and metastasis by disrupting trafficking and signaling pathways pivotal to cell proliferation and survival.
Acts as a GTPase-activating protein (GAP) for Rab GTPases, especially Rab4A and Rab5C, thereby modulating trafficking and signaling of receptors (such as EGFR); Inhibits viral gene transcription and replication by interacting with Rab5C; Promotes type I interferon responses during viral infection
