TBC1 domain family member 22A is a protein that functions as a GTPase-activating protein (GAP) for Rab family small GTPases, containing a conserved TBC domain that mediates its catalytic activity[2][1]. It is primarily localized to the Golgi apparatus and is involved in vesicle trafficking and protein-protein interactions, including binding to 14-3-3 proteins and forming homodimers[5][3]. TBC1D22A has shown elevated expression in ovarian cancer, where it is associated with poor prognosis, higher immune checkpoint expression, increased M2 macrophage infiltration, and greater chemoresistance to cisplatin and paclitaxel[2]. Its genetic alterations have been linked to several disorders, including epilepsy and schizophrenia, but it is not currently considered a direct therapeutic target (such as a receptor, enzyme, or transporter) and has no known drugs in clinical use targeting it directly[2][5].
