Overview

TBC1 domain family member 31 is a structural and molecular adapter protein encoded by the TBC1D31 gene and identified also as WDR67[3][6][7]. It is involved in the assembly of cilia, located at cellular structures such as the Golgi apparatus, centriolar satellite, and ciliary basal body[3][7]. TBC1D31 forms functional complexes with proteins like OFD1, potentially regulating their post-translational modifications including cAMP-dependent phosphorylation and ubiquitination[3]. In cancer biology, elevated TBC1D31 expression correlates with metabolic reprogramming (glycolytic phenotype) and is a prognostic biomarker in triple-negative breast cancer, suggesting a mechanistic link to aggressive cancer behavior[4]. There are currently no approved drugs directly targeting this protein in clinical settings.
