TBC1 domain family member 3E (TBC1D3E) is a member of the TBC (Tre-2/Bub2/Cdc16) domain-containing family with predicted GTPase activating protein (GAP) activity specific for RAB5, a small GTPase involved in endosome trafficking and membrane transport[2][5][6][9]. TBC1D3E is part of a rapidly evolving, human-specific gene family with multiple paralogs and copy-number variants, and does not act on RAB4 or RAB11[2][9]. Functionally, it regulates the release and molecular payload of extracellular vesicles (EVs) from immune and stromal cells, shaping inflammation, wound healing, and possibly tumor microenvironments by influencing the contents and biological activity of EVs[1]. Its expression can alter growth factor signaling and the innate immune response, and it has been implicated in cancer biology, especially in relation to its alternative names referencing prostate cancer[1][2]. TBC1D3E is not a classical druggable target but rather a regulatory protein influencing membrane trafficking pathways[2][6][9].
