Overview

TBC1 domain family member 3F (TBC1D3F) is a primate-specific member of the TBC1 domain family, implicated as a GTPase-activating protein (GAP) for RAB5, involved in the regulation and optimization of macropinocytosis via ARF6- and GGA3-dependent pathways[1][3]. The TBC1D3 family, consisting of closely related paralogs (A–H), is largely restricted to hominoids, with evidence of gene family expansion in humans[4]. TBC1D3F, like other TBC1D3 family members, is modestly expressed in most tissues but is elevated in the testis and brain[4]. It is overexpressed in several cancers—including kidney renal clear cell carcinoma—where its expression correlates with poor prognosis and immune infiltration, suggesting a role in tumor progression and as a potential prognostic biomarker[2][3]. While TBC1D3F is biologically significant, the TBC1D3 nomenclature is highly paralogous, and the specific functions or pathological roles of individual paralogs such as TBC1D3F remain incompletely differentiated in the literature[2][4].
