Overview

TBC1 domain family member 9 (TBC1D9) is a protein encoded by the TBC1D9 gene in humans and belongs to the TBC (Tre-2/Bub2/Cdc16) domain family, known for functioning as GTPase-activating proteins (GAPs) for Rab family small GTPases involved in membrane trafficking[5][7]. TBC1D9 contains two GRAM domains, a TBC domain, and an EF-hand motif, supporting roles in calcium binding, protein trafficking, and vesicle transport, particularly in endosomes and the Golgi[1][3]. It is implicated in the regulation of TBK1 activation during autophagy-related processes such as xenophagy and mitophagy in a calcium-dependent manner[1]. High TBC1D9 expression has been found in testicular germ cells and is involved in vesicular trafficking pathways related to Rab7 and Rab9[3]. In cancer biology, related TBC1 domain proteins are associated with metabolic phenotype regulation, but specific biomarker or drug targeting evidence for TBC1D9 is lacking[2][7].
