Overview

TBK1-binding protein 1 (TBKBP1) is a cytosolic adaptor protein that serves as a scaffold to recruit and coordinate signaling molecules, particularly the kinase TBK1, in pathways central to innate immunity and signal transduction[3]. TBKBP1 participates in the TNF/NF-κB pathway by linking TBK1 to various substrates and regulatory complexes, impacting processes such as interferon signaling, cellular stress response, and autophagy. As an adaptor, TBKBP1 does not possess enzymatic activity itself but is required for proper localization and activation of its binding partners. Disruption of TBKBP1 has implications in immune dysfunction, neurodegeneration, and cancer, especially in settings where TBK1 pathway modulation is critical for disease progression or therapy resistance[3].
