Overview

TEAD (TEA domain transcription factor) family proteins are a group of highly conserved transcription factors defined by their TEA/ATTS DNA-binding domain[1][5]. They serve as critical downstream effectors of the Hippo pathway, binding cofactors such as YAP, TAZ, and others to regulate gene transcription involved in growth, proliferation, and survival[2][6]. TEAD proteins play essential roles in organ development and homeostasis, and their dysregulation (most often via aberrant Hippo pathway signaling) has been implicated in tumorigenesis, stemness, and chemoresistance in various cancers[1][2][5]. Pharmacological inhibition of TEAD, especially disrupting their interaction with YAP/TAZ, is being explored as a novel anti-cancer approach[3][2][5].
