Teashirt zinc finger homeobox 2 (TSHZ2) is a nuclear transcription factor containing multiple zinc finger motifs and a homeobox domain[1][3]. It acts predominantly as a transcriptional repressor in association with C-terminal binding protein (CtBP), especially CtBP2[1][2]. TSHZ2 can inhibit oncogenic pathways by repressing downstream genes such as AEBP1 and CXCR4 and by inhibiting the transcription factor GLI1[1][2]. It interacts with several cell cycle regulators, including PRC1, KIF4A, and cyclin B1—implicating it in the control of mitosis and cytokinesis[2]. In normal tissues, TSHZ2 is expressed in the cell nucleus and is often found down-regulated in cancer, including breast and lung adenocarcinomas, where loss of expression associates with increased proliferation, invasion, and poor prognosis[1][2][3]. There are no known drugs that directly target TSHZ2, and its mechanisms of drug interaction, if any, have not been established. The gene and protein are well-characterized transcriptional repressors and tumor suppressors, but are not therapeutic targets for drugs as of current research, despite being critically relevant in cancer biology[1][2][3].
